It can take some practice. The guideline is to make each character distinctive in their own way. Through different camera angles, outfits, hair cuts, and personalities per character. Character 1 could be sitting, while character 2 is standing, Character 1 eats while character 2 twirls a pencil in their hand, etc.

One of the best actors to do this, is Ryan George, from the popular series Pitch Meetings. In the Behind the Scenes episode of the show he gives some great tips, and explains how to cut your audio so it sounds like two different people are talking in the same room at the same time.

Another great example is Seth Rogen portraying two different characters in the film An America Pickle.
